چکیده

INTRODUCTION There is continuing controversy surrounding force production and stability of muscle contraction on the descending limb of the force-length (F-L) relation. Many believe that force production on the descending limb is unstable. This notion is correct if the negative slope of the F-L relationship, which is obtained in a series of static, independent contractions, would also be negative for continuous dynamic contractions, such as stretching or shortening of the muscle. However, unstable behavior during muscle contraction has not been demonstrated by direct observation during dynamic muscle contraction, but has been inferred based on theoretical considerations or by measurement of average sarcomere length during contraction. In fact, there is evidence that sarcomeres may actually be stable on the descending limb of the F-L relation. While a muscle is stretched, force increases, thus establishing positive stiffness. If the residual force enhancement after stretch is larger than the force produced at the initial length, then force production in the muscle is stable. This has not been investigated using a whole muscle preparation, nor has it been discussed in the context of stability on the descending limb. Thus the purpose of this study was to investigate stability on the descending limb of the force length curve in an in situ muscle preparation.

A continuum-mechanical skeletal muscle model including actin-titin interaction predicts stable contractions on the descending limb of the force-length relation

Contractions on the descending limb of the total (active + passive) muscle force-length relationship (i. e. when muscle stiffness is negative) are expected to lead to vast half-sarcomere-length inhomogeneities. This is however not observed in experiments-vast half-sarcomere-length inhomogeneities can be absent in myofibrils contracting in this range, and initial inhomogeneities can even decreas...

Considerations on the history dependence of muscle contraction.

When a skeletal muscle that is actively producing force is shortened or stretched, the resulting steady-state isometric force after the dynamic phase is smaller or greater, respectively, than the purely isometric force obtained at the corresponding final length. The cross-bridge model of muscle contraction does not readily explain this history dependence of force production. The most accepted p...

Force enhancement following stretching of skeletal muscle: a new mechanism.

We investigated force enhancement following stretching in the in situ cat soleus muscle on the ascending and descending limb of the force-length relationship by varying the amount and speed of stretching and the frequency of activation (5 Hz, 30 Hz). There was a small but consistent (P<0.05) amount of force enhancement following muscle stretching on the ascending limb of the force-length relati...

Length dependence of active force production in skeletal muscle.

The sliding filament and cross-bridge theories of muscle contraction provide discrete predictions of the tetanic force-length relationship of skeletal muscle that have been tested experimentally. The active force generated by a maximally activated single fiber (with sarcomere length control) is maximal when the filament overlap is optimized and is proportionally decreased when overlap is dimini...
